Biographical notes:

John Penn and Richard Penn were proprietors of Pennsylvania at the time of writing to their brother Thomas. All three of them were sons of the founder of Pennsylvania, William Penn. The land dispute with Maryland mentioned in the letter was not resolved until 1760 when the border between Pennsylvania and Maryland was set between the 39 and 40 degrees mentioned in the letter.
