Middleton's writings consist of poems, essays, and addresses about Christianity, the fine arts, philosophical materialism, temperance, secession, fame, the U.S. Constitution, and other subjects, many of which were probably delivered to the students of the College of Charleston during his tenure there as professor and president. Correspondence includes letters (1845-1880) from Nathaniel Russell Middleton to his wife Anna and children concerning family matters, family financial affairs, religion, and other topics; letters (1856-1881) from Anna Middleton to Nathaniel Russell Middleton and her daughters Maria and Charlotte ("Lottie"); and letters (1868) from Nathaniel Russell Middleton, Jr. to his sister [Charlotte?] written while he was a student at the College of Charleston. Russell Middleton, Jr. discusses social outings, a "great firemen's parade" in Charleston, the appearance of the Battery, commencement exercises at the College of Charleston, and a prank played on the freshmen class by seniors. Miscellaneous items consist of a list (n.d.) of the names of educators at various institutions in South Carolina, and a page (1851) from an account book totalling expenses for tuition, shoes, and other items for Nathaniel Russell Middleton's son.

Papers consist of correspondence, financial records, writings, and other items. Correspondence (1820-1937) of the family of Nathaniel Russell Middleton (1810-1890) primarily concerns family and personal matters, but also business and College of Charleston affairs, the Civil War, and other matters. Correspondents include Nathaniel Russell Middleton, Nathaniel Russell Middleton, Jr., Anna E. De Wolf Middleton, Alicia H. Middleton, Maria Middleton (later Maria Doar), Charlotte H. Middleton (later Charlotte De Wolf), Arthur Middleton, Walter Izard Middleton, Henry Izard Middleton, Doar family members, and others. Letters (some typewritten transcriptions) are written from Charleston (S.C.), Bristol (R.I.), Stono Plantation (S.C.), Paris (France), and Rome (Italy). Financial records (1828-1900) include receipts for household and other expenses, invoices, bankbooks, bills, accounts, and a De Wolf family receipt book (1857-1870). Other items include the will (1930) of Alicia Hopton Middleton, a copy of the will (1889) of Anne Elizabeth De Wolf Middleton, floor plans of a house (n.d.), calling cards, clippings, invitations, and a pamphlet (ca. 1853) memorializing the death of four De Wolf family children. Writings include fragments of a typewritten manuscript of "Life in Carolina and New England during the Nineteenth Century" by Alicia Hopton Middleton (published 1929). Fragments are entitled "Reminiscences of Nathaniel Russell Middleton" and "Record of Nathaniel Russell Middleton, Jr." There are also typewritten copies of Middleton family correspondence (1844-1862) used in the book, and correspondence (1929-1930) concerning publication and sales of the book. Other writings include poetry collected or written by Middleton family members.

The bulk of the collection consists of Middleton and DeWolf family letters, many between family members in Bristol, R.I., and Charleston, S.C. In addition to standard family matters and the peculiarities of life in a family divided between the North and South, these letters and the other papers deal with such topics as Middleton's plantation, Bolton-on-the-Stono (apparently near Charleston), an 1849 slave insurrection, the College of Charleston, supply shortages during the Civil War, and selling rice and phosphate fertilizer during Reconstruction. Also included is "Narrative of his own Conversion" by Rev. John Joice, Darien, Ga., 1824.
