Born in New York City, Barry graduated from the United States Military Academy in 1838, 17th in his class of 45 cadets. He was commissioned a brevet second lieutenant in the 4th U.S. Artillery, transferring to the 2nd U.S. Artillery a few weeks later. He was stationed near the Canada–US border, then later took part in the Mexican–American, Seminole, and the Kansas-Missouri Border Wars.
He was the co-author of Instruction for Field Artillery (1860), along with William H. French and Henry J. Hunt.
Promoted to major of artillery shortly after the start of hostilities between the Union and the Confederacy, Barry served as Brig. Gen. Irvin McDowell's chief of artillery during the First Battle of Bull Run, where his position was overrun after mistaking advancing Confederates for retreating Union forces. Barry was appointed brigadier general by President Abraham Lincoln on August 20, 1861, to rank from August 20, 1861. President Lincoln submitted the nomination to the U.S. Senate on December 21, 1861 and the Senate confirmed it on March 17, 1862. Barry came up with the concept that became the U.S. Horse Artillery Brigade in the Army of the Potomac.
As chief of artillery under Maj. Gen. George B. McClellan, Barry organized ordnance for the Army of the Potomac and, during the Peninsula Campaign, later took part in the battles of Yorktown, Mechanicsville, Gaines' Mill, White Oak Swamp, and Malvern Hill.
After later supervising forts and ordnance surrounding Washington, D.C., Barry became chief of artillery under Maj. Gen. William T. Sherman, serving with him in Tennessee, the March to the Sea, and the Carolinas Campaign. On January 23, 1865, President Lincoln nominated Barry for appointment to the brevet grade of major general of volunteers, to rank from September 1, 1864, for his service in the Atlanta Campaign, and the U.S. Senate confirmed the appointment on February 14, 1865. Barry was mustered out of the volunteer force on January 15, 1866. On July 17, 1866, President Andrew Johnson nominated Barry for appointment to the brevet grade of major general in the Regular Army, to rank from March 13, 1865, and the U.S. Senate confirmed the appointment on July 23, 1866.
On December 11, 1865, Barry was appointed colonel in the 2nd U.S. Artillery, and was in command of the northern frontier during the Fenian raids of 1866. He served there until September 1867, and then commanded the artillery school of practice at Fort Monroe until March 1877, when he was appointed to the command at Fort McHenry, Baltimore, Maryland. During the labor riots of 1877 he rendered valuable service at Camden Station.
Barry died at Fort McHenry and is buried at Forest Lawn Cemetery in Buffalo, New York.

Griswold, discusses his desire to join the Ordnance Dept., and deprecates proposed candidate John Hills. Lynchburg druggest Howell Davies acknowledges an invoice from Seth Low & Co., of New York, 1833 January 11, and places an order for bottles syringes, snuff boxes and factory lamps. A letter, 1845 July 25, Rebecca Timmonds, a teacher in Rappanhannock County, Va., to Annie McLaughlin, Patapsco Female Institute, Ellicott's Mills, Md., describes life at her new school and comments on news from Patapsco. Gordon & Snapp, Winchester, orders 3 tons of pig iron from Joseph Marston & Company, Shenandoah Co., Va., 1852 February 5. James S. Griscom, Petersburg, Va., writes to Rene E. DeRussy [Old Point Comfort], 1854 May 29, regarding navigational improvements on the Appomattox River. [F. W. Risger?], Washington, D. C., writes to William G. King, New York, N.Y., 1858 May 28, concerning Capt. [J. L.]Cathcart's steam propeller boat and a Chesapeake and Ohio canal break. "Bee" writes to "Mollie" 1863 Sep 21, from behind federal lines conveying news of family and friends separated by war and describing a trip to Montgomery County, Md., and Washington, D. C. Letters written during the seige of Petersburg by John McGill, 1864 Aug 1 and [Mrs. A. J. Leavenworth?] 1864 Nov 17 discuss the purchase of property and scarce goods including shoes with a brief reference to the Battle of the Crater. [Pension agent?] Henri Bayard writes to his wife, 1888 Sep 4, describing pension day at the National Soldier A printed form letter from the State Central Committee of the Democratic Party of West Virginia is a call to action against the proposed 14th amendment and a solicitation for contributions. A handwritten list of signers and contributions is appended. Two letters from Edward D. Neill to Charles Deane, 1869 Sep 13 and Nov 12 on details in his History of the Virginia Company and expresses appreciation for Deane's colonial research. In a letter 1871 Mar 28 General William F. Barry requests Secretary of War W. W. Belknap to make an official visit to the Artillery School at Fort Monroe. [Pension agent?] Henri Bayard writes to his wife, 1888 Sep 4, describing pension day at the National Soldiers Home, Virginia, and being mistaken for her deceased father by a young child. Five letters from George N. Schuster to his sister Mary, Jul 1918-Jan 1919, humorously describe life in France and Germany, with a brief reference to the great flu epidemic. The collection also contains a document, signed by Harold Smyth, Wythe Chancery District Court Marshal, 1829 January 15, appointing Cornelius Baldwin Eskridge a deputy Marshall; a Baltimore Life Insurance Company application accomplished and signed by John H. Clack, with a forwarding letter to president J. J. Donaldson from agent Duncan Robinson,1835; two copies of an "Application of brothers and sisters for arrears of pay and other allowances," 1879 Jul 14, pertaining to the service of Albert Allmand on the U. S. Cyane and William H. Allmand on the U. S. Cumberland; and a printed postcard, 1909, describing and depicting Southern Seminary in Buena Vista, Va.
