The collection consists of letters, manuscripts and documents (all in Spanish) documenting the early history of Peru, from Franciso Pizarro's civil war with Almagro to the pacification of the country by La Gasca. It includes material pertaining to the Catholic Church in Peru, missionaries, the conquest of Chile, Indians of Peru, and Indian slavery. There are materials regarding the cities of Arequipa, Cuzco, Lima, and Quito. Of note are letters and decrees of Charles V and Philip II, letters of the Pizarros, reports of La Gasca, and Pedro de Valdivia's account of the conquest of Chile.

Collection consists of letters, documents and transcripts relating to the discovery, conquest and colonization of Brazil, Ecuador, Mexico, Paraguay, Peru, Venezuela, the Moluccas, and the Philippines; and to the Casa de Contratación and the Council of the Indies. Correspondents include the kings of Spain and their emissaries in the New World. Transcripts are from the 1780s and mostly in the hand of Antonio de Ugina. They appear largely to duplicate other transcripts in the Muñoz Collection at the Biblioteca de la Real Academia de la Historia in Madrid. In some cases, however, they are believed to be the only surviving copies of documents.

Transcripts, translations, and summaries of documents and letters relating primarily to civil and ecclesiastical administration of the Central American provinces during the sixteenth century, to the development of navigation routes and the exploration of the Pacific in that vicinity, and to the character and treatment of the Indians. Most of these volumes bear annotations signed by Buckingham Smith, 1856-1857, and sometimes also by Martín Fernández de Navarrete or his assistant, certifying the location of the original. Record relating to the transfer of the remainder of Balboa's fleet to González Dávila; royal agreement with the pilot, Andrés Niño, for discoveries in the Pacific; and Gonzálex Dávila's report of March 6, 1524, on his discoveries in Nicaragua. Excerpts from Volumes 66, 67, and 69 (new numbering) of the Muñoz Collection, Academia Real de la Historia, Madrid. These are transcripts and summaries of letters and documents addressed chiefly to Emperor Charles I of Spain or to the Council of the Indies by officials, friars, and Indians of the provinces subject to the Audiencia de los Confines, namely, Guatemala, Honduras, Nicaragua, Yucatan, and Chiapa. They relate to the civil and ecclesiastical administration of these provinces, particularly to treatment of Indians, abolition of the encomienda system, the work of Dominican and Franciscan friars, administration of Alonso de Zurita, the expeditions of Pedro de la Gasca, activities of Francisco Pizarro in Peru, and the survey conducted by Diego Ramírez.

A collection of autographs of Latin American patriots and Hispanic loyalists living in the Viceroyalty of Perú during the 19th century. The collection provides a valuable look into daily life in the viceroyalty. Documents to and from military leaders on both sides of the War for Independence describe day to day events of the war (requests for supplies, mess, prisoner and hospital lists, etc.). Among the items by patriots represented in the collection are: a decree by Simón Bolivar, dated 1825, setting up a school system that "la debe igualmente a todos...individuos."; a decree by Tomas de Heres creating the coat-of-arms of Perú, and the heraldry and wording for silver and gold coins of the new government; a list by José Medina of the survivors of the "Vendedores" squadron after the battle of Ayachucho; a letter of José de San Martín to Francisco de Paula Otero; a letter by Antonio José de Sucre to Manuel Martínez de Aparicio; and a letter by José de La Mar to Joaquín de la Pezuela. Among the loyalists represented are: José Fernando de Abascal y Souza, Fernando VII, José Antonio de Areche, Gerónimo de Marrón y Lombera, José de Mendizábal e Imaz, Joaquín de la Pezuela, José Manuel Goyeneche, and Manuel Quimper. Also in the collection are two letters by Secretary of State William E. Seward, and two 16th century documents signed by the "conquistadores" Francisco Pizarro and Juan de la Torre.
