The bulk of the collection consists of correspondence to Earle from his colleagues, dealing with issues pertinent to their interests in the field of mental illness. There are letters about patients, case notes, and drafts for reports. There are also letters from psychiatrists in Europe and a few items of personal correspondence from members of his family. One of his innovations in the treatment of patients was to offer lectures to them on various literary and cultural topics. Included in this collection are a number of those lectures as well as copies of some of his articles. There are twenty-eight diaries, 1835-1892, in which Earle wrote brief daily entries, accounts, addresses, and genealogical notes. There are also several volumes of medical and travel notebooks and one octavo volume of copies of letters to his family written from Europe, Cuba, and Washington, D.C., 1837-1864. Earle worked on the genealogy of his family for over fifty years and there are several folders of notes and correspondence gathered in preparation for his book. There is also some biographical material on Earle by Franklin Benjamin Sanborn (1831-1917) who wrote the "official" biography of him. At the beginning of the collection there are two manuscripts pertaining to Earle's father and his development of carding machines for the textile industry. One of the items is a letter, April 1804, from Eleazar Smith ( - ) of Walpole, Mass., describing improvements for what appears to be a carding machine.

A print of Child is also present. Correspondents include John Sullivan Dwight, Pliny Earle, James Thomas Fields, Anthony Philip Heinrich, Thomas Wentworth Higginson, Harriet Jacobson, Oliver Johnson, Frances Locke, Lucretia Mott, Samuel Stillman Osgood, Israel Post, and Richard Henry Stoddard.

The earliest letters in this collection are between the elder Pliny Earle and his brothers Jonah (1765-1846), Henry (1774-1837), and Timothy (1778-1819) regarding the wool card manufacturing business. The correspondence of John Milton Earle and his family begins with letters between Earle and Sarah Hussey in the period of their courtship, and these letters contain much discussion of reading and Quakerism. Later letters include many to and from Earle's children, particularly Ann H. Earle, although many members of the family are correspondents and recipients. Other correspondents include Lucy Chase (1822-1909) of Worcester. Although focusing on family and local matters, these letters cover a very wide range of topics. Numerous letters between John Milton Earle and his wife Sarah date to the period after their marriage, as she frequently visited Nantucket. These letters contain much news of events in Worcester and Nantucket. The papers of the younger Pliny Earle include a series of diaries from the years 1831 to 1833 as well as travel and medical notes from his visits to Europe and Turkey in 1838-1839. There are also genealogical notes on the Earle family and a "description of Elizabeth B. Chase's Spiritual Communications" dated 1852. Miscellaneous materials include a group of subscriptions for the Quaker Meeting, Leicester, Mass., 1790's; genealogical notes compiled by John Milton Earle, and penmanship exercise books and an account book of Sarah Hussey. There is also a small amount of material concerning the Mulberry Grove Boarding School, a school for young women in Leicester conducted from 1827 to 1839 by John Milton and Pliny Earle's sisters Sarah (1800-1834), Lucy (1805-1887), and Eliza (1807-1846).
