H. H. Dukes obtained a B.S. at Clemson College in 1915 and D.V.M. at Iowa State College in 1918 where he was a Teaching Fellow in Veterinary Physiology 1915-1918. 1918-1920, he was Assistant Professor of Veterinary Science at Clemson College and Assistant State Veterinarian of South Carolina; 1920-1921, Veterinary Practice and Health Officer, Greer, South Carolina; 1921-1932, Instructor and Assistant Professor of Veterinary Physiology and Assistant Professor of Veterinary Research (Physiology), Iowa State College.

He came to Cornell in 1932, teaching and conducting research in veterinary physiology including electro-cardiology, ruminant physiology, poultry metabolism, and porcine respiratory reflexes. In 1947 he was asked to teach a series of sessions at Cornell on the demonstration technique he had developed. From these sessions which "taught teachers" grew the American Society of Veterinary Physiologists and Pharmacologists. In 1953 as a visiting professor in the Cultural Division of the Brazilian Ministry of Foreign Affairs he lectured at veterinary, medical, philosophical and agricultural colleges throughout that country. That same year he also lectured at the Veterinary School in Lima, Peru. After long service to Cornell he retired in 1960 as a professor emeritus. He was a member of the American Veterinary Medical Association, the National Research Council, the Southern Tier Veterinary Medial Association and the State Veterinary Medical Society as well as being a fellow of the American Association for the Advancement of Science and member of New York Academy of Sciences, American Physiological Society, Society for Experimental Biology and Medicine, State Society for Medical Research and five honorary societies: Sigma Xi, Phi Kappa Phi, Phi Zeta Phi, Phi Lambda Upsilon and Alpha Psi.
