Luke Milbourne was educated at Cambridge and ordained in 1670. A supporter of Henry Sacheverell, he was appointed lecturer of St. Leonard, Shoreditch in 1688 and rector of St. Ethelburga, Bishopsgate, London, in 1704. He published 31 single sermons, several tracts, and Notes on Dryden's Virgil (1698), in which he attacked Dryden's skill as a translator.
